Inhibitionof the Lectin-induced Mitogenic Response of Thymocytes by Glycolipids1
نویسندگان
چکیده
Exogenous gangliosides at concentrations found in Se muminhibit the concanavalin A(Con A) induced mitogenic response of mouse thymocytes. Of four gangliosides tested, the tnisialoganglioside, °T1'was the most potent inhibitor. Cemamides, cemebmosides, and sialic acid were not inhibitory at any concentration tested. The inhibition by gangliosides was not due to interference with Con A binding as shown by direct binding studies with [3HJacetyI-Con A nor was it due to a nonspecific killing eftect. Thymocytes exposed to a ganglioside concentration 5 times that required to inhibit mitogenesis were still capable of excluding trypan blue up to 44 hr after ganglioside addition. Furthermore, ganglio side inhibition could be reversed by washing the cells 4 hr after addition of the glycolipid. A productive interaction with Con A occurs in the presence of ganglioside as shown by a Con A-induced increase in carbohydrate metabolism. However, uridine and thymidine incorporation are inhibited by the presence of ganglioside. Complete inhibition could be achieved if the glycolipid were added as late as 24 to 28 hr after the Con A in a 48-hr mitogenic assay. The results are discussed in light of recent findings that elevated levels of gangliosides are found in the sera of tumor-bearing animals, and it is suggested that gangliosides shed by tumor cells could be involved in the generalized immuno suppression observed in such animals.

Cell-mediated mitogenic response induced by leukoagglutinin and Lens culinaris lectin in mouse lymphocytes
The proliferative response of mouse lymphocytes to syngeneic cellular stimulation upon membrane modification with lectins was studied. Brief pretreatment of stimulator cells (mitomycin-C-treated spleen cells) followed by mixed culture with syngeneic cortisone-resistant thymocytes resulted in a significant proliferative response in the thymocytes. This effect was not due to a soluble mediator an...
متن کاملCell-mediated Mitogenic Response Induced by Leukoagglutinin
This work was undertaken to characterize the response of mouse lymphocytes to a syngeneic stimulus after surface modification by lectins. As previously reported (1), cortisone-resistant thymocytes (CRT) 1 manifest a high proliferative response to syngeneic mitomycin-C-treated spleen cells (Mito-Sp) 2-3 days after the initiation of culture when either responder or stimulator is briefly pretreate...
متن کاملInhibition of mitogenic stimulant-induced activation of thymocytes with zinc tetrakis-(N-methyl-4'-pyridyl) porphyrinato.
Zinc porphyrins have anti-inflammatory and anti-allergic properties. The objective of the present study was to characterize the mechanism of zinc tetrakis-(N-methyl-4'-pyridyl) porphyrinato (ZnTMPyP) immune modulation by investigating its effects on the proliferative activity during thymocyte stimulation with mitogenic factors and the molecular events mediating thymocyte proliferation. The resu...
متن کاملA novel activation pathway for mature thymocytes. Costimulation of CD2 (T,p50) and CD28 (T,p44) induces autocrine interleukin 2/interleukin 2 receptor-mediated cell proliferation
Prior studies have shown that thymocytes, unlike peripheral T cells, do not proliferate in response to mitogenic combinations of anti-CD2 mAbs. The present study demonstrated that stimulation by a mitogenic anti-CD2 combination (9-1 plus 9.6) with anti-CD28 induced vigorous thymocyte proliferation in the absence of exogenous IL-2. This thymocyte proliferation was IL-2 dependent as shown by the ...
متن کامل[The pharmacological effects of disodium ethane-1-hydroxy-1, 1-diphosphonate (EHDP) on the function of immunocompetent cells. 1. The mitogenic response of thymocytes or spleen cells of young rats injected with EHDP].
The effect of in vivo administration of disodium ethan-l-hydroxy-1.1-diphosphonate (EHDP) on the mitogenic response of thymocytes or spleen cells were studied in young rats. When rats (2-3 weeks of age) were injected with EHDP (10mg/kg/day) subcutaneously for more than 6 days or injected once with EHDP (100mg/kg) subcutaneously or intraperitoneally, lipopolysaccharide-induced proliferative resp...
